AAK1 also known as adaptor-associated kinase 1 is a protein kinase with a molecular weight of 88 kDa. It plays a significant role in the regulation of clathrin-mediated endocytosis. AAK1 phosphorylates the mu2 subunit of the AP2 complex which is critical for the recruitment of clathrin to membranes. This protein is expressed in various tissues with high levels noted in the brain and spinal cord. AAK1 also participates in the modulation of neurotransmitter receptors and synaptic vesicle function.
Biological function summary

AAK1 functions as part of the AP2 clathrin adaptor complex which is essential for the internalization of cargo at the plasma membrane. Its activity is important for maintaining proper cellular uptake of nutrients receptors and signaling molecules. In neurons AAK1 regulates the cycle of synaptic vesicles impacting neurotransmission by influencing vesicle release and recycling. It has been suggested that AAK1 also contributes to the modulation of dendritic spine morphology through its kinase activity.

Pathways

AAK1 participates in endocytosis and intracellular trafficking pathways. It is directly involved in the clathrin-mediated endocytic route interacting with other proteins such as clathrin and dynamin. This association with clathrin provides a mechanism for cargo selection and membrane curvature linking AAK1 to the broader system of intracellular transport. In addition AAK1 connects to MAPK signaling through potential cross-talk mechanisms highlighting its role in cellular signaling regulation.

AAK1 has connections to neurodegeneration and cancer. In diseases like Alzheimer's abnormal function or expression of AAK1 can affect synaptic activity potentially exacerbating memory loss and cognitive decline. AAK1 also shows altered expression patterns in some cancers suggesting a role in oncogenic pathways. Its interaction with proteins like ALK and Ret may influence cell migration and proliferation reflecting AAK1's impact on disease states.

Regulates clathrin-mediated endocytosis by phosphorylating the AP2M1/mu2 subunit of the adaptor protein complex 2 (AP-2) which ensures high affinity binding of AP-2 to cargo membrane proteins during the initial stages of endocytosis (PubMed : 11877457, PubMed : 11877461, PubMed : 12952931, PubMed : 14617351, PubMed : 17494869, PubMed : 25653444).
