Supplementary information

AANAT also known as aralkylamine N-acetyltransferase or serotonin N-acetyltransferase is an enzyme with a molecular mass of approximately 23.3 kDa. This enzyme catalyzes the transfer of an acetyl group from acetyl-CoA to serotonin forming N-acetylserotonin. AANAT expression is mainly found in the pineal gland where it acts as an important player in melatonin biosynthesis. The function of AANAT as a regulatory point in this biosynthesis contributes to the role of melatonin in circadian rhythms and sleep regulation.
Biological function summary

AANAT has a critical role in regulating physiological processes through the production of melatonin. It operates as part of the intramolecular complex that facilitates its enzymatic activity in the pineal gland. By converting serotonin into N-acetylserotonin which further converts to melatonin AANAT modulates the synthesis of melatonin an indoleamine with effects on sleep-wake cycles and seasonal reproductive rhythms in mammals. This regulatory activity reflects how AANAT influences the hormonal signaling pathways in response to light and dark cycles.

Pathways

AANAT is integral to the melatonin synthesis pathway and interacts closely in serotonin metabolism. It tightly regulates the conversion of serotonin to melatonin in the pineal gland especially during nocturnal periods. One key protein related to AANAT in this pathway is hydroxyindole O-methyltransferase (HIOMT) which converts N-acetylserotonin to melatonin. The modulation of melatonin levels impacts diverse biological pathways including circadian rhythm regulation and neuroendocrine signaling.

AANAT plays a significant role in conditions like seasonal affective disorder (SAD) and sleep disturbances. Alterations in AANAT expression or function can affect melatonin levels leading to these disorders. Additionally disruptions in the enzymatic activity of AANAT may have interactions with serotonin influencing its availability and potentially linking it to disorders like depression. Understanding AANAT's function allows researchers to explore therapeutic strategies targeting melatonin pathways in managing these conditions.
