Supplementary information

ABCA7 also known as ATP-binding cassette sub-family A member 7 functions as a transmembrane protein that plays a role in lipid transport and homeostasis. It belongs to the large ABC transporter family characterized by its ability to bind and hydrolyze ATP to transport various substrates across cellular membranes. The protein's mass is approximately 220 kDa. ABCA7 is mostly expressed in the brain and immune cells such as macrophages and microglia where it influences cellular lipid composition and related processes.
Biological function summary

The ABCA7 protein participates in lipid metabolism and immune response regulation. It is not part of a larger protein complex but functions in concert with other proteins to modulate processes such as phospholipid efflux and phagocytosis. Its role in macrophage-mediated lipid efflux affects the uptake and processing of lipids in cellular contexts which can impact inflammation and cellular signaling pathways connected to immunity.

Pathways

ABCA7 contributes significantly to lipid metabolism and immune response pathways. In lipid metabolism it shows functional interactions with proteins like ABCA1 which also participates in cholesterol efflux and HDL formation. In immune pathways it affects macrophage activity by regulating the phagocytic capability of these cells linking it to processes that influence inflammation and the immune system's ability to clear amyloid-beta peptides.

Mutations or alterations in ABCA7 expression have connections to Alzheimer's disease and atherosclerosis. In Alzheimer's disease ABCA7 alters the amyloid-beta clearance process in the brain an important feature of the disease's pathology with implications pointing to interaction with proteins like APOE a known risk factor. In atherosclerosis ABCA7 influences lipid processing and inflammation in arterial macrophages affecting the buildup of atherosclerotic plaques and related cardiovascular conditions.

Target data

Catalyzes the translocation of specific phospholipids from the cytoplasmic to the extracellular/lumenal leaflet of membrane coupled to the hydrolysis of ATP (PubMed : 24097981). Transports preferentially phosphatidylserine over phosphatidylcholine (PubMed : 24097981). Plays a role in lipid homeostasis and macrophage-mediated phagocytosis (PubMed : 12917409, PubMed : 12925201, PubMed : 14570867, PubMed : 14592415). Binds APOA1 and may function in apolipoprotein-mediated phospholipid efflux from cells (PubMed : 12917409, PubMed : 14570867, PubMed : 14592415). May also mediate cholesterol efflux (PubMed : 14570867). May regulate cellular ceramide homeostasis during keratinocyte differentiation (PubMed : 12925201). Involved in lipid raft organization and CD1D localization on thymocytes and antigen-presenting cells, which plays an important role in natural killer T-cell development and activation (By similarity). Plays a role in phagocytosis of apoptotic cells by macrophages (By similarity). Macrophage phagocytosis is stimulated by APOA1 or APOA2, probably by stabilization of ABCA7 (By similarity). Also involved in phagocytic clearance of amyloid-beta by microglia cells and macrophages (By similarity). Further limits amyloid-beta production by playing a role in the regulation of amyloid-beta A4 precursor protein (APP) endocytosis and/or processing (PubMed : 26260791). Amyloid-beta is the main component of amyloid plaques found in the brains of Alzheimer patients (PubMed : 26260791).
