Supplementary information

ABCB10 also known as ATP-binding cassette sub-family B member 10 plays a role as a transport protein involved in the mitochondrial export of molecules. It is a member of the ABC (ATP-binding cassette) transporter family characterized by its ability to move various substrates across membranes using ATP hydrolysis. ABCB10 has a molecular mass of approximately 81 kDa and is expressed mainly in the mitochondria of cells with significant levels in tissues with high metabolic activity like the heart and liver.
Biological function summary

ABCB10 contributes to important cellular functions by participating in heme synthesis. It interacts with other mitochondrial proteins forming complexes that facilitate the import of iron and export of heme precursors. This interaction ensures the maintenance of proper heme levels which are necessary for diverse cellular processes including respiration and detoxification pathways. Its activity is not isolated but connected to broader protein networks within the mitochondrial intermembrane space.

Pathways

ABCB10 participates in the heme biosynthesis pathway which is essential for the production of hemoproteins including hemoglobin and cytochromes. Within this context ABCB10 works closely with proteins such as ferrochelatase which catalyzes the final step in heme production. Another related pathway is oxidative phosphorylation where ABCB10 indirectly sustains energy production by ensuring the availability of heme for cytochrome assembly which is pivotal for electron transport chain function.

ABCB10 plays a significant role in conditions related to mitochondrial dysfunction such as sideroblastic anemia and cardiovascular diseases. Mutations affecting ABCB10 can disrupt normal heme synthesis impacting red blood cell production and leading to anemia. Additionally improper function of ABCB10 influences iron homeostasis which associates with proteins like ALAS2 (aminolevulinate synthase 2) further contributing to erythropoietic disorders and highlighting its role in iron-related pathologies.

Target data

ATP-dependent transporter located in the mitochondrial inner membrane that catalyzes the export of biliverdin from the mitochondrial matrix, and plays a crucial role in hemoglobin synthesis and antioxidative stress (PubMed : 22085049, PubMed : 28315685, PubMed : 28808058, PubMed : 34011630, PubMed : 37041204). Participates in the early step of the heme biosynthetic process during insertion of iron into protoporphyrin IX (PPIX) (PubMed : 22085049, PubMed : 28808058). Involved in the stabilization of the iron transporter mitoferrin-1/SLC25A37 (By similarity). In addition may be involved in mitochondrial unfolded protein response (UPRmt) signaling pathway, although ABCB10 probably does not participate in peptide export from mitochondria (PubMed : 28315685).
