ATP-dependent transporter of the ATP-binding cassette (ABC) family that actively extrudes physiological compounds, and xenobiotics from cells. Lipophilic anion transporter that mediates ATP-dependent transport of glucuronide conjugates such as estradiol-17-beta-o-glucuronide and GSH conjugates such as leukotriene C4 (LTC4) (PubMed:12527806, PubMed:15256465). May contribute to regulate the transport of organic compounds in testes across the blood-testis-barrier (Probable). Mediates multidrug resistance (MDR) in cancer cells by preventing the intracellular accumulation of certain antitumor drugs, such as, docetaxel and paclitaxel (PubMed:15256465, PubMed:23087055). Does not transport glycocholic acid, taurocholic acid, MTX, folic acid, cAMP, or cGMP (PubMed:12527806).

Activity summary

The ABCC10 protein also known as Multidrug Resistance Protein 7 (MRP7) functions as an ATP-binding cassette transporter. It contributes to multidrug resistance by extruding a variety of xenobiotics and physiological substrates out of cells. The ABCC10 protein has a mass of about 171 kDa. Researchers have found ABCC10 expression in various tissues including the liver lung and testis suggesting its diverse role in different cell types.

Biological function summary

This transporter plays critical roles in cellular detoxification processes. ABCC10 is part of the larger ATP-binding cassette (ABC) transporter family which includes other transporters like ABCC1 and ABCC2. By actively transporting drugs and organic anions across cellular membranes ABCC10 helps regulate the intracellular concentrations of these compounds and maintain cellular homeostasis.

Pathways

ABCC10 participates in the detoxification and xenobiotic metabolism pathways interacting with enzymes such as cytochrome P450s. The protein contributes to the resistance against various chemotherapy drugs by exporting them out of cells impacting the drug metabolism pathway. This function is related to other ABC transporters like ABCC1 which similarly influence drug clearance and pharmacokinetics.

Associated diseases and disorders

ABCC10 has been implicated in cancer chemotherapy resistance. Many studies have noted that overexpression of ABCC10 correlates with reduced drug accumulation in cancer cells resulting in chemotherapy failure. Its function is also connected to the protein P-glycoprotein another drug transporter that plays a similar role in multidrug resistance highlighting the complex network of transporters involved in therapeutic resistance.
