We have systematically measured KD (the equilibrium dissociation constant between the antibody and its antigen), of more than 840 recombinant antibodies to assess not only their individual KD values but also to see the average affinity of antibody. Based on the comparison with published literature values for mouse monoclonal antibodies, Recombinant antibodies appear to be on average 1-2 order of magnitude higher affinity.

Supplementary information

ABI2 also known as Abl-Interactor 2 is a protein encoded by the ABI2 gene. It has a molecular mass of approximately 53 kDa. ABI2 functions as a regulator of actin cytoskeleton dynamics and participates in processes involving cell motility and morphology. This protein is mainly expressed in neuronal tissues and is found in the cytoplasm suggesting its involvement in intracellular signaling pathways where actin rearrangement is important.
Biological function summary

ABI2 impacts cellular functions by interacting with the WAVE (WASP-family verprolin-homologous) complex. This interaction facilitates the regulation of actin polymerization important for processes such as cell migration and synaptic plasticity. ABI2 through the WAVE complex plays a role in the formation of lamellipodia cellular protrusions important for movement. This regulation ensures that cells respond effectively to external stimuli by reorganizing their cytoskeleton.

Pathways

The protein contributes significantly to the regulation of the Rac1 signaling pathway important for cytoskeletal remodeling. ABI2 works closely with proteins such as NCK proteins and phosphoinositides that are part of this pathway. Furthermore ABI2 interacts with the PAK family of kinases influencing downstream signaling that controls cell cycle progression and survival. ABI2’s role in these pathways highlights its importance in maintaining cellular architecture and dynamics.

ABI2 is implicated in neurological conditions particularly Alzheimer's disease and schizophrenia. Its interaction with other proteins like APP (amyloid precursor protein) suggests a role in synaptic dysfunction seen in these disorders. ABI2’s involvement in actin dynamics connects it to synaptic defects that may contribute to cognitive impairments in Alzheimer's disease. Understanding ABI2’s function and interactions offers insights into therapeutic targets for these neurological conditions.

Target data

Regulator of actin cytoskeleton dynamics underlying cell motility and adhesion. Functions as a component of the WAVE complex, which activates actin nucleating machinery Arp2/3 to drive lamellipodia formation (PubMed : 21107423). Acts as a regulator and substrate of nonreceptor tyrosine kinases ABL1 and ABL2 involved in processes linked to cell growth and differentiation. Positively regulates ABL1-mediated phosphorylation of ENAH, which is required for proper polymerization of nucleated actin filaments at the leading edge (PubMed : 10498863, PubMed : 7590236, PubMed : 8649853). Contributes to the regulation of actin assembly at the tips of neuron projections. In particular, controls dendritic spine morphogenesis and may promote dendritic spine specification toward large mushroom-type spines known as repositories of memory in the brain (By similarity). In hippocampal neurons, may mediate actin-dependent BDNF-NTRK2 early endocytic trafficking that triggers dendrite outgrowth (By similarity). Participates in ocular lens morphogenesis, likely by regulating lamellipodia-driven adherens junction formation at the epithelial cell-secondary lens fiber interface (By similarity). Also required for nascent adherens junction assembly in epithelial cells (PubMed : 15572692).
