Binds to zinc finger protein TNFAIP3 and inhibits NF-kappa-B activation induced by tumor necrosis factor, Toll-like receptor 4 (TLR4), interleukin-1 and 12-O-tetradecanoylphorbol-13-acetate. Overexpression inhibits NF-kappa-B-dependent gene expression in response to lipopolysaccharide at a level downstream of TRAF6 and upstream of IKBKB. NF-kappa-B inhibition is independent of TNFAIP3 binding.

ABIN3 regulates immune response and participates in controlling inflammation. It acts together with other components to form signaling complexes. This complex assembly plays a role in dampening excessive inflammatory signals. By regulating components of innate immune responses ABIN3 helps maintain immune homeostasis. Its interaction is significant in controlling the overall activity within signaling hubs that manage inflammation.
ABIN3 also known as TNIP3 is a protein with a molecular mass of approximately 68 kDa. It is widely expressed in various tissues notably in the immune system. ABIN3 acts as an important mediator in modulating cellular responses to external stress signals. It interacts with several signaling pathways modulating functions at the cellular level. ABIN3's action significantly regulates immune responses by binding to and inhibiting specific proteins involved in inflammation.
ABIN3 involves itself in critical inflammatory pathways such as the NF-κB pathway. This protein modulates NF-κB activation by hindering its activity ensuring proper balance in immune signaling. ABIN3 connects with proteins like A20 acting together to prevent overactivation that could lead to inflammation. Such modulation is central in managing cellular defenses and preventing inappropriate tissue damage.
Researchers link ABIN3 to conditions like inflammatory bowel disease (IBD) and rheumatoid arthritis. In these diseases ABIN3's role in moderating immune response becomes relevant as dysfunctions here can lead to excessive inflammation.
