ab154396 was shown to react with ACAT1 in western blot. The band observed in the knockout cell line ab269621 (knockout cell lysate ab272431) lane below 45 kDa is likely to represent a truncated form. This has not been investigated further. Membranes were blocked in 3 % milk in TBS-T (0.1 % Tween®) before incubation with ab154396 and ab8245 (Mouse anti-GAPDH antibody [6C5]) overnight at 4°C at a 1 in 500 dilution and a 1 in 20000 dilution respectively. Blots were incubated with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed (ab216776) secondary antibodies at 1 in 20000 dilution for 1 h at room temperature before imaging.

Biological function summary
ACAT1 plays a central role in cellular cholesterol homeostasis and in preventing cholesterol-induced cytotoxicity by converting free cholesterol into inert cholesteryl esters for storage. It functions as a homotetramer suggesting that it works as a part of a larger protein complex. ACAT1 influences lipid accumulation within cells and mediates the regulation of cholesterol levels which is vital for maintaining membrane integrity and cellular function.
