Supplementary information

ADAM15 also known as Metargidin is a member of the ADAM (a disintegrin and metalloproteinase) family of proteins. It has a mass of approximately 92 kDa. This protein is a transmembrane protein with metalloprotease activity which means it can cleave other transmembrane or extracellular matrix proteins. ADAM15 is expressed in various tissues including heart lung liver and kidney and is notably present in cells of the immune system and certain cancerous tissues.
Biological function summary

ADAM15 influences cell adhesion migration and signaling pathways. It operates as part of both integrin-mediated and growth factor-mediated signaling complexes. This protein modulates interactions between integrins and the extracellular matrix impacting cellular movement and communication. By processing and regulating these interactions ADAM15 plays a role in tissue remodeling and inflammatory responses.

Pathways

ADAM15 interacts with proteins in the integrin signaling and EGFR (epidermal growth factor receptor) pathways. This involvement makes it a critical component in cell signaling linked to proliferation differentiation and survival. Specifically ADAM15 influences integrin-related proteins such as β1-integrin and is implicated in modulating the signaling activity of EGFR. These interactions illustrate its role in the regulation of cellular dynamics.

ADAM15 has been connected to cancer and arthritis. Its overexpression has been observed in some cancer types including breast and prostate cancer where it affects tumor growth and metastasis potential. Additionally ADAM15 contributes to the pathology of arthritis by regulating inflammation and cartilage degradation. Through these processes ADAM15 works alongside proteins like matrix metalloproteinases in the disease mechanisms.

Target data

Active metalloproteinase with gelatinolytic and collagenolytic activity. Plays a role in the wound healing process. Mediates both heterotypic intraepithelial cell/T-cell interactions and homotypic T-cell aggregation. Inhibits beta-1 integrin-mediated cell adhesion and migration of airway smooth muscle cells. Suppresses cell motility on or towards fibronectin possibly by driving alpha-v/beta-1 integrin (ITAGV-ITGB1) cell surface expression via ERK1/2 inactivation. Cleaves E-cadherin in response to growth factor deprivation. Plays a role in glomerular cell migration. Plays a role in pathological neovascularization. May play a role in cartilage remodeling. May be proteolytically processed, during sperm epididymal maturation and the acrosome reaction. May play a role in sperm-egg binding through its disintegrin domain.
