ICC/IF image of ab28302 stained PC12 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1% BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilize the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab28302, 1 μg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43 μM.

The bands observed at 75 kDa could potentially be a cleaved form of ADAM23 due to the presence of a signal peptide and propeptide.

ADAM23 also known as A Disintegrin And Metalloprotease 23 is a member of the ADAM family of proteases. This protein has a mass of approximately 95 kDa. ADAM23 is expressed in several tissues but it has a significant presence in the brain. The protein mechanically functions by engaging in protease activities though it is uniquely categorized due to its role beyond traditional proteolytic functions. It interacts with surrounding cell-matrix elements through disintegrin domains.
Biological function summary

ADAM23 plays a role in processes such as cell-cell adhesion and signal transduction. It aids in the regulation of synapse formation and maintenance in neuronal cells which is important for proper neural development and function. ADAM23 is not generally considered a part of large protein complexes but collaborates in the cellular environment impacting spatial cellular frameworks and growth signaling cascades.

Pathways

Multiple mechanisms rely on ADAM23 especially those involved in cell signaling. The protein interacts with integrin pathways influencing cellular adhesion and migration dynamics. Furthermore it works closely with other members of the ADAM family such as ADAM22 in modulating signaling pathways critical for neuronal function and transmission.

ADAM23 has shown connections to neurological conditions notably epilepsy and some types of cancer.
