Supplementary information

ADAMTS10 also referred to as A Disintegrin and Metalloproteinase with Thrombospondin Motifs 10 is a member of the ADAMTS family. This protein has an approximate molecular mass of 110 kDa. The ADAMTS10 protein functions mainly as a metalloprotease involved in the cleavage of extracellular matrix components affecting tissue remodeling and development processes. It is expressed in various tissues including the eyes and heart suggesting its broad role in different physiological contexts.
Biological function summary

ADAMTS10 plays a significant role in maintaining the structure of fibrillin-rich microfibrils which are essential components of the extracellular matrix. It is part of a larger complex involved in cell signaling and tissue integrity. The interaction with fibrillin suggests its involvement in connective tissue maintenance and elastogenesis which is important for normal cellular functions in eye and cardiac tissue.

Pathways

ADAMTS10 provides functionality within fibrillin and microfibril assembly pathways impacting processes like tissue morphogenesis. It associates closely with other matrix proteins like fibrillin-1 and ADAMTSL proteins in these pathways influencing the structural integrity and biomechanical properties of tissues. Additionally its regulation can affect Notch signaling pathways linking to cellular growth and differentiation processes.

ADAMTS10 mutations are linked to Weill-Marchesani syndrome a connective tissue disorder characterized by eye heart and skeletal abnormalities. The protein's activity also associates with glaucoma a condition involving elevated intraocular pressure leading to optic nerve damage. Researchers have noted a connection with LTBP2 (latent-transforming growth factor beta-binding protein 2) in these conditions suggesting that mutations or dysfunctions in this pathway can disrupt fibrillin functions and lead to disease pathogenesis.
