ADAMTS12 also recognized as A Disintegrin and Metalloproteinase with Thrombospondin Motifs 12 is a member of the ADAMTS family. It is a secreted enzyme with a mass of approximately 204 kDa. This protease is involved in extracellular matrix remodelling. The expression of ADAMTS12 occurs mainly in placental tissue and at lower levels in various other tissues including the lung and heart. It plays a role in processing protein substrates in the extracellular environment.
Biological function summary

The interactions with other matrix components allow ADAMTS12 to mediate matrix organization and degradation. It does not form part of a larger protein complex on its own but interacts with extracellular matrix proteins like aggrecan and versican. These interactions facilitate the breakdown of large proteoglycans aiding tissue morphogenesis and repair.

Pathways

ADAMTS12 participates in the extracellular matrix organization and degradation pathways. These pathways involve proteins such as aggrecanase and versicanase which ADAMTS12 closely relates to through its proteolytic activity. It plays a significant role in matrix remodelling which is essential for cellular migration and differentiation processes.

Alterations in ADAMTS12 expression link to osteoarthritis and cancer. In osteoarthritis increased ADAMTS12 activity contributes to cartilage degradation. In cancer its expression modifies cellular environments influencing tumor progression. It also interacts with matrix proteins like aggrecan in osteoarthritis and demonstrates associations with cancer-related proteins contributing to disease pathology.
