ADAMTS4 also known as aggrecanase-1 is a protein responsible for extracellular matrix degradation. It weighs approximately 90 kDa. This metalloproteinase is expressed in various tissues including cartilage and synovial tissue. It cleaves aggrecan a core protein of the proteoglycan into fragments impacting the integrity and function of the cartilage matrix. It is secreted as a latent enzyme and undergoes activation in the extracellular space.
ADAMTS4 influences tissue remodeling through the breakdown of cartilage. It contributes to the disassembly of aggrecan and other matrix components influencing extracellular matrix turnover. As part of a group of similar enzymes it plays a role in proteolytic processes important for normal tissue maintenance and repair. Its actions affect the balance between matrix synthesis and degradation which is important for maintaining tissue homeostasis.
ADAMTS4 participates in pathways such as the inflammation-related cytokine signaling and the degradation of the extracellular matrix. This protein interacts closely with ADAMTS5 contributing to matrix degradation and modulation of inflammatory processes. In these pathways the balanced activity of ADAMTS4 is essential for proper cartilage function and the resolution of inflammation in tissues.
ADAMTS4 is notably linked to osteoarthritis and rheumatoid arthritis. These conditions involve excessive cartilage breakdown where ADAMTS4 and ADAMTS5 contribute significantly to disease progression through aggrecan degradation. Understanding the activity and regulation of ADAMTS4 offers insights into potential therapeutic strategies for managing cartilage degeneration and inflammatory joint diseases.
