Metalloproteinase that plays an important role in connective tissue organization, development, inflammation and cell migration. Extracellular matrix (ECM) degrading enzyme that show proteolytic activity toward the hyalectan group of chondroitin sulfate proteoglycans (CSPGs) including ACAN, VCAN, BCAN and NCAN (PubMed:16133547, PubMed:18992360). Cleavage within the hyalectans occurs at Glu-Xaa recognition motifs. Plays a role in embryonic development, including limb and cardiac morphogenesis, and skeletal muscle development through its VCAN remodeling properties. Cleaves VCAN in the pericellular matrix surrounding myoblasts, facilitating myoblast contact and fusion which is required for skeletal muscle development and regeneration (By similarity). Participates in development of brown adipose tissue and browning of white adipose tissue (By similarity). Plays an important role for T-lymphocyte migration from draining lymph nodes following viral infection.

ADAMTS5 also known as aggrecanase-2 is a member of the ADAMTS (a disintegrin and metalloproteinase with thrombospondin motifs) family. Its approximate mass is 120 kDa. This enzyme functions primarily as a metalloprotease cleaving proteoglycans such as aggrecan thereby playing a significant role in the degradation of extracellular matrix components. ADAMTS5 is widely expressed in tissues such as cartilage. It is integral for maintaining tissue homeostasis and flexibility within the joint structures.
ADAMTS5 significantly contributes to the dynamic remodeling of the extracellular matrix. It operates without forming complexes with other proteins and functions independently. By cleaving aggrecan it regulates cartilage integrity balancing synthesis and breakdown of matrix constituents. This balance is essential for facilitating normal cartilage function and enables joints to resist load and stress.
Current understanding highlights that ADAMTS5 is critical within pathways like cartilage catabolism and inflammation response. ADAMTS5 activity initiates extracellular matrix remodeling by interacting with proteins such as aggrecan and collagen II. It plays a role in precise regulatory processes that manage cartilage homeostasis and maintains stability in structural integrity through its catabolic actions.
ADAMTS5 has strong links to osteoarthritis and rheumatoid arthritis. Pathological overexpression or dysregulated activity of ADAMTS5 can accelerate cartilage degradation within these conditions.
