Immunocytochemistry - Anti-ADAMTS9 antibody (AB32565)

Immunofluorescence analysis of ADAMTS9 in SH-SY5Y cells using ab32565. The cells were fixed with 4% paraformaldehyde, permeabilized with 0.1% Triton™ X-100, and blocked with 1% BSA. The cells were labeled with ab32565 at 2 μg/mL in 0.1% BSA and incubated for 3 hours at room temperature followed by a Alexa Fluor® 488 Goat anti-Rabbit IgG (H+L) Secondary Antibody at 1/2000 dilution for 45 minutes at room temperature (Panel a : green). Nuclei (Panel b : blue) were stained with with DAPI. F-actin (Panel c : red) was stained with Alexa Fluor® 555 Rhodamine Phalloidin at 1/300. (Panel d) represents the merged image showing localization in extracellular matrix. (Panel e) shows the no primary antibody control. The images were captured at 60X magnification.

The ADAMTS9 protein also known as A Disintegrin and Metalloproteinase with Thrombospondin motifs 9 plays a mechanical role as a metalloproteinase enzyme. It features a molecular weight of approximately 193 kDa. ADAMTS9 is expressed in various tissues including the heart pancreas and kidney among others. This protein undergoes post-translational modifications that impact its mechanical functions offering unique regulation mechanisms for tissue remodeling and extracellular matrix degradation.
Biological function summary

ADAMTS9 influences processes associated with maintaining extracellular matrix integrity and remodeling. It participates in the cleavage of proteoglycans hence modulating cellular environments. This protein does not typically form large complexes but it interacts with substrates that are part of the complex networks found in connective tissues. Its enzymatic activity is subject to intervention by specific inhibitors which can significantly affect tissue homeostasis.
