ADCK4 also known as aarF domain containing kinase 4 is a protein kinase with a molecular weight of approximately 49 kDa. It mainly exists in the mitochondria where it plays a significant role in cellular energy production. Expression of ADCK4 appears highest in the kidney and to a lesser extent in other tissues. As a kinase its mechanical function depends on phosphorylating various substrates which is necessary for the maintenance of mitochondrial function.
Biological function summary

ADCK4 participates in the biosynthesis of coenzyme Q10 a vital component of the electron transport chain. This protein forms part of a larger complex known as the coenzyme Q biosynthesis complex where it helps maintain cellular energy and metabolic balance. It interacts closely with other proteins involved in this pathway to regulate coenzyme Q10 levels which affects multiple cellular processes including ATP production.

Pathways

ADCK4 significantly integrates into the mitochondrial respiratory chain pathway and the broader metabolic energy pathway. It aids in the synthesis of coenzyme Q10 alongside proteins like COQ6 and COQ7 directly impacting the efficiency of energy production in mitochondria. Mutations or dysfunctions in ADCK4 correlate with nephrotic syndrome and coenzyme Q10 deficiency. Target data

Atypical kinase involved in the biosynthesis of coenzyme Q, also named ubiquinone, an essential lipid-soluble electron transporter for aerobic cellular respiration (PubMed : 24270420, PubMed : 36302899, PubMed : 38425362). Its substrate specificity is still unclear : may act as a protein kinase that mediates phosphorylation of COQ3 (PubMed : 38425362). According to other reports, acts as a small molecule kinase, possibly a lipid kinase that phosphorylates a prenyl lipid in the ubiquinone biosynthesis pathway, as suggested by its ability to bind coenzyme Q lipid intermediates (By similarity). However, the small molecule kinase activity was not confirmed by another publication (PubMed : 38425362). Required for podocyte migration (PubMed : 24270420).
