Supplementary information

The ADCY10 protein also known as soluble adenylate cyclase (sAC) functions as an enzyme that produces cyclic adenosine monophosphate (cAMP) from ATP. Weighing approximately 187 kDa it acts in a unique way compared to other adenylate cyclases due to its solubility in the cytosol rather than being membrane-bound. ADCY10 expresses in various tissues with abundant expression observed in testis and sperm cells. Its expression pattern and physicochemical properties underpin its distinctiveness among the cyclase family.
Biological function summary

ADCY10 plays a significant role in generating cAMP in response to bicarbonate and calcium ions which are key to sperm motility and fertilization processes. This catalytic activity heavily influences metabolic activities and sperm functions cementing its role in reproductive biology. Although it does not typically form a multi-protein complex ADCY10's interaction at a molecular level allows it to regulate vital cellular activities important to maintaining proper cellular responses to extracellular signals.

Pathways

ADCY10 serves in the cAMP signaling pathway an essential cascade that transduces extracellular signals into cellular responses. This pathway's crosstalk with others such as the calcium signaling pathway involves proteins like protein kinase A (PKA) which ADCY10 indirectly activates. Through these pathways ADCY10 impacts various physiological processes signifying its pivotal role in cellular communication and response.

ADCY10 has been linked to male infertility primarily due to its role in sperm activation and motility. Abnormalities in ADCY10 function might contribute to impaired sperm motility. Additionally ADCY10 has associations with other sperm-specific proteins such as CatSper channels which are important for proper sperm function. Disruptions in these proteins' interactions could further elucidate ADCY10's involvement in reproductive health issues.

Target data

Catalyzes the formation of the signaling molecule cAMP (PubMed : 12609998, PubMed : 15659711, PubMed : 24567411, PubMed : 24616449, PubMed : 25040695). May function as sensor that mediates responses to changes in cellular bicarbonate and CO(2) levels (PubMed : 15659711, PubMed : 17591988). Has a critical role in mammalian spermatogenesis by producing the cAMP which regulates cAMP-responsive nuclear factors indispensable for sperm maturation in the epididymis. Induces capacitation, the maturational process that sperm undergo prior to fertilization (By similarity). Involved in ciliary beat regulation (PubMed : 17591988).
