ADCY2 also known as Adenylate Cyclase 2 is an enzyme involved in the synthesis of cyclic adenosine monophosphate (cAMP) from ATP. ADCY2 with a molecular mass of approximately 140 kDa plays an important role in signal transduction. This protein is highly expressed in the brain particularly in areas such as the cortex and the hippocampus. As part of the adenylate cyclase family ADCY2 is one of several isoforms that share similar functions in different tissues.
Biological function summary

ADCY2 contributes to various cellular processes by converting ATP to cAMP an important secondary messenger in cells. It forms part of the larger signalosome complex and interacts with G-protein coupled receptors. By regulating cAMP levels ADCY2 influences processes such as neurotransmitter release and long-term memory formation. It also modulates the activity of protein kinase A impacting several downstream signaling pathways.

Pathways

ADCY2 is integral to G-protein coupled receptor signaling and the cAMP signaling pathway. This pathway plays a fundamental role in transmitting extracellular signals to intracellular responses. ADCY2's activity is closely linked with other proteins like G-proteins and phosphodiesterases which help regulate cAMP levels and downstream cellular responses. In some pathways ADCY2 interacts synergistically with the protein CREB which orchestrates gene transcription in response to cAMP levels.

ADCY2 has been associated with neurological conditions such as depression and schizophrenia. Altered expression or function of ADCY2 can lead to imbalances in cAMP signaling which affect mood regulation and cognitive functions. The protein CREB which ADCY2 influences is also implicated in mood disorders and cognitive impairments.
