In Western blot, ab220408 was shown to bind specifically to Adenine Nucleotide Translocator 1 + 2/ANT 1 + 2. First, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 3 % milk in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4°C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed (ab216776) at 1/20000 dilution.

Biological function summary
ANT 2 and ANT 1 contribute to maintaining mitochondrial function and integrity. They form part of the larger mitochondrial permeability transition pore complex which plays a role in energy metabolism and apoptosis. By regulating ADP/ATP exchange ANT 2 and ANT 1 ensure efficient energy production aligning cellular energy consumption with metabolic demands. Dysfunctions in their activity can result in impaired ATP synthesis affecting overall cellular viability and function.
Pathways
ANT 2 and ANT 1 participate significantly in critical processes like oxidative phosphorylation and the apoptosis pathway. In oxidative phosphorylation these proteins interact with the F1F0 ATP synthase complex to facilitate ATP production. Within the apoptosis pathway involvement in the opening of the mitochondrial transition pore can trigger programmed cell death. Connections with proteins such as cytochrome c occur in these pathways highlighting their role in energy regulation and cell survival.
