Immunocytochemistry/ Immunofluorescence - Anti-Adenovirus Type 5 antibody (AB6982)

ab6982 staining Adenovirus type 5 in human HeLa cells by Immunocytochemistry/ Immunofluorescence. The cells were fixed in paraformaldehyde, permeabilised in 0.1% Triton and then blocked using 0.1% BSA for 16 hours at 4°C. Samples were then incubated with primary antibody at 1/800 for 1 hour at 37°C. The secondary antibody used was a donkey anti-rabbit IgG conjugated to Alexa Fluor® 546 (red) used at a 1/1000 dilution.

Adenovirus Type 5 also known as Ad5 is a non-enveloped virus with a double-stranded DNA genome. It is a member of the adenoviridae family which exhibits an icosahedral capsid structure and an approximate mass of 150 MDa. Adenoviruses including Type 5 target various human tissues predominantly the epithelial cells in the respiratory gastrointestinal and urinary tracts. They utilize CAR (Coxsackie and Adenovirus Receptor) for cell entry showcasing their specific interaction with host cells.
Biological function summary

Adenoviruses like Type 5 are associated with several intracellular processes and response mechanisms. Typically they are not part of a larger biological complex but interact dynamically with the host cell machinery. During the infection process Ad5 manipulates host replication and transcription pathways to facilitate its own replication subverting normal cellular controls. This action often results in the inhibition of host cell DNA synthesis while encouraging viral replication.

Pathways

The adenovirus Type 5 engages significantly with the cellular pathways regulating apoptosis and immune response modulation. It interacts with p53 and E1A proteins which are essential influencers of cell cycle control and apoptosis. Through these interactions Ad5 can modulate the host's apoptotic pathways ensuring the virus can replicate efficiently while evading immune surveillance and promoting viral persistence.

Adenovirus Type 5 is most notably linked to respiratory illnesses and conjunctivitis. In respiratory infections Ad5 exploits the immune modulation pathways resulting in inflammation and symptoms of the common cold. It can associate with other proteins like the E3 region proteins helping evade host immune responses and leading to prolonged infection or more severe symptoms. In the context of ocular infections such as conjunctivitis similar mechanisms apply whereby the adenovirus disregards normal immune clearance perpetuating the infection cycle.
