Supplementary information

ADHFE1 also known as alcohol dehydrogenase iron-containing 1 is a mitochondrial protein with a molecular mass of approximately 62 kDa. It is expressed in various tissues with higher levels in the liver kidney and heart. Functioning as a member of the iron-containing alcohol dehydrogenase family ADHFE1 catalyzes the conversion of alcohols into aldehydes an essential step in the metabolic pathway involving alcohols.
Biological function summary

The enzyme facilitates important redox reactions that contribute to various metabolic processes. ADHFE1 is not part of a larger complex but acts independently in its role of oxidizing alcohols to aldehydes. It impacts metabolic regulation by modulating the levels of substrates and products involved in energy production and detoxification which is significant for maintaining cellular homeostasis.

Pathways

ADHFE1 plays an important role in the metabolic network of energy conversion and alcohol detoxification. It is integrated into the glycolytic pathway and interacts with other proteins like G6PD and LDHA which function in glucose metabolism and lactate production respectively. These interactions support cellular energy production and manage the fate of glucose-derived metabolites.

ADHFE1 involvement relates to metabolic disorders such as alcohol dependence and non-alcoholic fatty liver disease (NAFLD). In alcohol dependence ADHFE1's activity influences the concentration of acetaldehyde which may contribute to the disease's pathology. Additionally its expression and interaction with proteins like ADH1C and ALDH2 are relevant in the context of liver diseases impacting the metabolic balance and detoxification processes in the liver.

Target data

Catalyzes the cofactor-independent reversible oxidation of gamma-hydroxybutyrate (GHB) to succinic semialdehyde (SSA) coupled to reduction of 2-ketoglutarate (2-KG) to D-2-hydroxyglutarate (D-2-HG). D,L-3-hydroxyisobutyrate and L-3-hydroxybutyrate (L-3-OHB) are also substrates for HOT with 10-fold lower activities.
