Immunocytochemistry/ Immunofluorescence - Anti-ADIPOR1 antibody (AB70362)
ab70362 staining ADIPOR1 in HepG2 cells. The cells were fixed with 4% paraformaldehyde (10 min), permeabilized with 0.1% PBS-Tween for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1%PBS-Tween for 1h. The cells were then incubated overnight at 4°C with ab70362 at 5µg/ml and ab7291, Mouse monoclonal [DM1A] to alpha Tubulin - Loading Control. Cells were then incubated with ab150081, Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (Alexa Fluor® 488), pre-adsorbed at 1/1000 dilution (shown in green) and ab150120, Goat polyclonal Secondary Antibody to Mouse IgG - H&L (Alexa Fluor® 594), pre-adsorbed at 1/1000 dilution (shown in pseudocolour magenta). Nuclear DNA was labelled with DAPI (shown in blue). Image was acquired with a high-content analyser (Operetta CLS, Perkin Elmer) and a maximum intensity projection of confocal sections is shown.

Immunocytochemistry/ Immunofluorescence - Anti-ADIPOR1 antibody (AB70362)
ab70362 staining ADIPOR1 in HepG2 cells treated with sodium 4-phenylbutyrate (ab141253), by ICC/IF. Increase of aADIPOR1 expression correlates with increased concentration of sodium 4-phenylbutyrate, as described in literature.
The cells were incubated at 37°C for 6 hours in media containing different concentrations of ab141253 (sodium 4-phenylbutyrate) in DMSO, fixed with 4% formaldehyde for 10 minutes at room temperature and blocked with PBS containing 10% goat serum, 0.3 M glycine, 1% BSA and 0.1% tween for 2h at room temperature. Staining of the treated cells with ab70362 (5 μg/ml) was performed overnight at 4°C in PBS containing 1% BSA and 0.1% tween. A DyLight® 488 anti-rabbit polyclonal antibody (ab96899) at 1/250 dilution was used as the secondary antibody. Nuclei were counterstained with DAPI and are shown in blue.

Immunocytochemistry/ Immunofluorescence - Anti-ADIPOR1 antibody (AB70362)
ab70362 staining ADIPOR1 in HepG2 cells treated with valproic acid, sodium salt (ab120745), by ICC/IF. Increased cytoplasmatic staining of ADIPOR1 correlates with increased concentration of valproic acid, as described in literature.
The cells were incubated at 37øC for 24h in media containing different concentrations of ab120745 (valproic acid) in DMSO, fixed with 4% formaldehyde for 10 minutes at room temperature and blocked with PBS containing 10% goat serum, 0.3 M glycine, 1% BSA and 0.1% tween for 2h at room temperature. Staining of the treated cells with ab70362 (5 μg/ml) was performed overnight at 4øC in PBS containing 1% BSA and 0.1% tween. A DyLight 488 goat anti-rabbit polyclonal antibody (ab96899) at 1/250 dilution was used as the secondary antibody. Nuclei were counterstained with DAPI and are shown in blue.

Biological function summary
ADIPOR1 facilitates the metabolic actions of adiponectin by increasing insulin sensitivity and promoting fatty acid oxidation. The receptor does not function alone; it operates in coordination with another isoform adiponectin receptor 2 (ADIPOR2) forming a complex to exert its full biological effects. This dual receptor system supports the regulation of energy homeostasis and inflammation influencing metabolic rate and improving lipid profiles.
Pathways
ADIPOR1 plays a critical role in the AMP-activated protein kinase (AMPK) pathway and the peroxisome proliferator-activated receptor alpha (PPAR-α) signaling pathway. These pathways are important for cellular energy balance and lipid metabolism. AMPK activation by ADIPOR1 leads to enhanced glucose uptake and fatty acid oxidation while interaction within the PPAR-α pathway regulates gene expression related to lipid metabolism. The activity of ADIPOR1 is intricately tied to other metabolic proteins like leptin and insulin thereby linking with these pathways.
