Supplementary information

ADM2 also known as adrenomedullin 2 or intermedin is a peptide with various roles in the body. This peptide consists of 47 amino acids and usually has a molecular mass of about 5.1 kDa. You can find ADM2 expression in several tissues including the heart kidney and brain where it carries out several physiological functions. In these tissues ADM2 acts through binding to the calcitonin receptor-like receptor (CLR) in the presence of receptor activity-modifying proteins (RAMPs).
Biological function summary

The function of ADM2 centers on its role as a vasoactive peptide. ADM2 possesses vasodilatory effects helping regulate blood pressure and fluid balance. It forms part of a receptor complex that includes CLR and specific RAMPs allowing it to exert its effects efficiently. This interaction modulates vascular tone and cardiac output contributing to overall cardiovascular health.

Pathways

ADM2 is active in the G protein-coupled receptor signaling pathway. Its effects are closely linked to the cyclic AMP (cAMP) signaling pathway an essential pathway for numerous physiological responses. ADM2 interacts with proteins such as CLR and RAMPs facilitating cAMP production and eliciting downstream effects that include vasodilation and other protective cardiovascular effects.

Researchers have noted the involvement of ADM2 in cardiovascular diseases and hypertension. Its regulatory effect on blood vessels links it closely with conditions such as hypertension where increased ADM2 activity may represent a protective mechanism. ADM2's relationship with proteins like CLR and RAMPs suggests therapeutic potential for managing conditions involving vascular dysfunction.

Target data

Intermedin/ADM2 is a peptide hormone that plays a role as physiological regulator of gastrointestinal and cardiovascular bioactivities mediated by the CALCRL-RAMPs receptor complexes (PubMed : 14615490). Activates the cAMP-dependent pathway through interaction with CALCRL-RAMP3 receptor complex (PubMed : 14615490).
