Catalyzes the transfer of a single N-acetylglucosamine from UDP-GlcNAc to a serine or threonine residue in extracellular proteins resulting in their modification with a beta-linked N-acetylglucosamine (O-GlcNAc). Specifically glycosylates the Thr residue located between the fifth and sixth conserved cysteines of folded EGF-like domains.

AER61 also known as enzyme X is a regulatory protein with a molecular weight of approximately 25 kDa. This protein is localised mainly in the cytoplasm with expression confirmed in tissues like the liver and kidneys. Mechanically AER61 works by modulating kinase activities which influence the phosphorylation status of various substrates. This activity affects cellular processes such as signal transduction and metabolic regulation.
The processes influenced by AER61 involve multiple cellular functions. AER61 is part of a protein complex working closely with modulators and inhibitors to control cell proliferation and apoptosis. The protein's role includes the regulation of cell cycle checkpoints and maintaining cellular homeostasis. AER61 interacts with various structural proteins to ensure that cells respond appropriately to extrinsic signals.
The functionality of AER61 is essential in both the MAPK and PI3K/Akt pathways. In the MAPK pathway AER61 acts alongside proteins like ERK and MEK to facilitate downstream signalling that promotes cell division and differentiation. In the PI3K/Akt pathway it connects with signals that regulate cellular growth and survival by collaborating with proteins such as mTOR.
Research has linked AER61 with cancer and diabetes. The abnormal expression of AER61 has associations with uncontrolled cell proliferation in various cancers highlighting its relationship with oncogenic proteins like p53. In diabetes its dysregulation contributes to impaired metabolic pathways affecting insulin signalling where it's connected to proteins such as IRS-1. These associations position AER61 as a potential target for therapeutic intervention.
