Immunohistochemical analysis of paraffin-embedded human colon carcinoma tissue labeling AGO4 with ab259821 at 1/100 (5.02 μg/ml) dilution followed by ready to use Rabbit specific IHC polymer detection kit HRP/DAB (ab209101). Cytoplasmic staining on human colon carcinoma (PMID : 20146808). The section was incubated with ab259821 for 30 mins at room temperature. The immunostaining was performed on a Leica Biosystems BOND® RX instrument. Counterstained with Hematoxylin.

AGO4 was immunoprecipitated from 0.35 mg THP-1 (human monocytic leukemia monocyte), whole cell lysate 10 μg with ab259821 at 1/30 dilution (2μg in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab259821 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP)(ab131366) was used at 1/5000 dilution.

Biological function summary
AGO4 participates in small RNA pathways. It is part of the RNA-induced silencing complex and works closely with DICER and TRBP to process and guide small RNA molecules. This process affects cellular mRNA levels by reducing the translation of target mRNA molecules helping regulate gene expression post-transcriptionally.
Pathways
AGO4 functions in the RNA interference and gene silencing by microRNAs pathways affecting post-transcriptional gene regulation. AGO4 interacts with proteins like DICER1 and other Argonaute proteins such as AGO1-3 within these pathways aiding in the effective downregulation of specific mRNA targets.
