Paraffin-embedded human pancreatic cancer tissue stained for AGPS with ab236621 at a 1/500 dilution in immunohistochemical analysis.
After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

NIH/3T3 (Mouse embryo fibroblast cell line) cells stained for AGPS using ab236621 at a dilution of 1/166 in ICC/IF.
The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal goat serum. The cells were then incubated with the primary antibody overnight at 4°C. Secondary used is an Alexa-Fluor®488-conjugated Goat Anti-Rabbit IgG (H+L).

Biological function summary
AGPS functions as an important component in the synthesis of ether lipids specifically plasmalogens which play a role in membrane structure and signaling. Plasmalogens are phospholipids that are part of cellular membranes and influence membrane fluidity and dynamics. AGPS operates as a sole enzyme in the initial step of de novo plasmalogen biosynthesis engaging with other enzymes to complete this lipid complex. Its activity ensures the proper formation of plasmalogens which are particularly abundant in the heart and nervous tissues.
Pathways
AGPS plays a central role in the ether lipid biosynthesis pathway and interacts with proteins involved in lipid metabolism. Plasmalogen biosynthesis is a critical pathway where AGPS catalyzes the first step acting as a precursor in the synthesis of several ether lipids.
