HepG2 (human liver hepatocellular carcinoma cell line) cells stained for AGXT2 using ab237622 at a dilution of 1/233 in ICC/IF.

The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal goat serum. The cells were then incubated with the primary antibody overnight at 4°C. Secondary used is an Alexa-Fluor®488-conjugated Goat Anti-Rabbit IgG (H+L). Counterstained with DAPI.

AGXT2 also known as alanine--glyoxylate aminotransferase 2 is an enzyme with approximately 56 kDa mass. The enzyme is expressed mainly in the liver kidney and heart. AGXT2 catalyzes the transamination reaction between alanine and glyoxylate to produce pyruvate and glycine. It is critical for the cellular detoxification processes and the regulation of amino acid metabolism.
Biological function summary

This enzyme contributes to the conversion of glyoxylate therefore preventing its accumulation which can be harmful. AGXT2 is not part of a larger protein complex but it works closely with other enzyme systems involved in amino acid metabolism. Its activity maintains the balance between glycolate and glycine levels within the body which is essential for normal cellular function.

Pathways

AGXT2 functions in both the glyoxylate and dicarboxylate metabolism pathway as well as the broader amino acid metabolism pathways. Within these pathways its role connects it to other related proteins such as Glyoxylate reductase/hydroxypyruvate reductase (GRHPR) and Lactate dehydrogenase. These interactions contribute to maintaining homeostasis in intermediary metabolism.

AGXT2 abnormalities relate closely to primary hyperoxaluria type 2 and certain cardiovascular diseases. Mutations or deficiencies in AGXT2 can result in the overproduction of oxalate leading to kidney stone formation and renal damage. Additionally reduced AGXT2 activity might influence homocysteine levels potentially linking it to cardiovascular disorders. This relationship places AGXT2 in connection with other proteins such as cystathionine beta-synthase in the context of homocysteine metabolism.

Catalyzes the conversion of glyoxylate to glycine using alanine as the amino donor (By similarity). Catalyzes metabolism of not L- but the D-isomer of D-beta-aminoisobutyric acid to generate 2-methyl-3-oxopropanoate and alanine (PubMed : 24586340). Catalyzes the transfer of the amino group from beta-alanine to pyruvate to yield L-alanine and 3-oxopropanoate (By similarity). Can metabolize NG-monomethyl-L-arginine (NMMA), asymmetric NG,NG-dimethyl-L-arginine (ADMA) and symmetric NG,N'G-dimethyl-L-arginine (SDMA) (PubMed : 20018850, PubMed : 23023372).
