Supplementary information

AIP1 also known as ASK1-interacting protein-1 is a multifunctional protein involved in the regulation of apoptosis and inflammation. It possesses a mass of approximately 90 kDa and is widely expressed in various tissues including the heart lung liver and skeletal muscle. This protein plays an important role in cell signaling pathways serving as an adapter protein in stress-related pathways. It binds to and regulates the function of apoptosis signal-regulating kinase 1 (ASK1) influencing cellular stress responses and survival decisions.
Biological function summary

The actions of AIP1 extend beyond simple interaction with ASK1. AIP1 is involved in the modulation of inflammatory processes and oxidative stress impacting cell damage and repair. It functions as a part of a complex signaling network where it influences cellular responses to external stress by interacting with other proteins. Its interaction with signaling molecules modulates pathways that control apoptosis making it an important regulator of cell fate.

Pathways

AIP1 is engaged in the MAPK signaling and JNK pathways where it regulates responses to cellular stress and apoptosis. AIP1 interacts significantly with proteins like tumor necrosis factor (TNF) receptor-associated factor 2 (TRAF2) which are critical in these pathways. Through these interactions AIP1 influences the balance between cell death and survival under stress conditions contributing to cellular homeostasis.

The involvement of AIP1 in apoptosis and inflammation links it to conditions such as cancer and cardiovascular disease. Its regulation of stress-response pathways implicates alterations in AIP1 function with the progression of these diseases. For instance abnormal expression of AIP1 can impact the regulatory role of ASK1 in cancerous cell survival. Furthermore dysregulated AIP1 activity might disrupt normal apoptotic processes and exacerbate inflammatory responses contributing to cardiovascular pathology through interactions with related proteins such as TRAF2.

Target data

Seems to act as a scaffold molecule at synaptic junctions by assembling neurotransmitter receptors and cell adhesion proteins (By similarity). Plays a role in nerve growth factor (NGF)-induced recruitment of RAPGEF2 to late endosomes and neurite outgrowth (By similarity). May play a role in regulating activin-mediated signaling in neuronal cells (By similarity). Enhances the ability of PTEN to suppress AKT1 activation (PubMed : 10760291). Plays a role in receptor-mediated clathrin-dependent endocytosis which is required for ciliogenesis (By similarity).
