Immunohistochemical analysis of paraffin-embedded Mouse thymus tissue labelling AIRE with ab270972 at 1/100 (4.52 ug/ml) followed by a ready to use LeicaDS9800 (Bond™ Polymer Refine Detection). Nuclear staining on mouse thymus medulla (PMID : 18214467, PMID : 22448160). The section was incubated with ab270972 for 30 mins at room temperature. The immunostaining was performed on a Leica Biosystems BOND® RX instrument Counterstained with Hematoxylin.
Secondary antibody only control : Secondary antibody is a ready to use LeicaDS9800 (Bond™ Polymer Refine Detection).
Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0, epitope retrieval solution2) for 20 mins

Biological function summary
AIRE functions to maintain central tolerance in the immune system. It acts as a part of a transcriptional complex that regulates genes involved in immune tolerance. This complex controls the expression of tissue-specific antigens (TSAs) important for "educating" T cells in the thymus to distinguish between self and non-self antigens. By controlling this mechanism AIRE helps to lower the risk of the body attacking its own tissues.
Pathways
AIRE contributes significantly to the pathway of immune tolerance and T cell selection within the thymus. It interacts with several other proteins involved in these pathways including the tumor necrosis factor receptor superfamily member RANK influencing the maturation of mTECs. Additionally AIRE interacts with proteins like CHD4 and a range of histone-modifying enzymes to modulate gene expression more broadly within the context of epigenetic regulation.
