Supplementary information

AKAP12 also known as A-kinase anchoring protein 12 or Gravin is a scaffolding protein that orchestrates the spatial and temporal coordination of signaling pathways. It has a molecular mass of approximately 150 kilodaltons and is primarily expressed in various tissues including the heart brain and skeletal muscle. AKAP12 functions by anchoring protein kinase A (PKA) and other signaling molecules to specific cellular compartments enabling efficient signal transduction. The protein’s structural adaptability allows for the assembly of multiprotein complexes which are important for its mechanical role in cells.
Biological function summary

The functions of AKAP12 extend beyond simple scaffolding. It interacts with other components to form macromolecular complexes that regulate cellular processes such as cytoskeletal dynamics cell cycle progression and signal transduction. By binding to PKA and other kinases such as protein kinase C (PKC) AKAP12 modulates phosphorylation events essential for maintaining cellular homeostasis. It also plays roles in regulating gene expression and cellular responses to external stimuli influencing processes like cell migration and proliferation.

Pathways

AKAP12 significantly contributes to the regulation of the MAPK/ERK and PI3K/Akt signaling pathways. Within these pathways AKAP12 acts by organizing multiple signaling proteins into functional networks influencing cell growth and survival. It closely interacts with components like Raf and PKB/Akt facilitating effective signal relay. These interactions ensure tight regulation of key cellular activities including growth differentiation and survival which are pivotal for normal cellular function.

Research implicates AKAP12 in cancer progression and cardiovascular diseases. It appears to act as a tumor suppressor in certain cancers where its downregulation or loss correlates with poor prognosis and increased metastatic potential. Its interaction with proteins like integrins further suggests a role in promoting or inhibiting cancer cell migration and invasion. Additionally AKAP12's involvement in cardiovascular diseases relates to its function in vascular smooth muscle cell migration and the maintenance of endothelial barrier function potentially interacting with proteins such as endothelial nitric oxide synthase (eNOS).
