AKAP14 also known as A-kinase anchor protein 14 plays an essential role in anchoring protein kinase A (PKA) to specific intracellular compartments. It weighs approximately 100 kDa. Researchers found it primarily in the centrosome and mitochondria. The protein enables the localization and functions of PKA which modulates various cellular processes. Through its capacity to tether PKA AKAP14 facilitates targeted phosphorylation of substrates impacting cellular signaling dynamics and structural integrity.
Biological function summary

AKAP14 helps organize and maintain the structural framework of cells by anchoring PKA in cellular compartments. As part of a larger protein complex AKAP14 interacts with other structural proteins facilitating their assembly and function. This function is important for maintaining centrosome integrity and proper mitochondrial function. The anchoring partners of AKAP14 ensure precise PKA activity where required affecting cell cycle regulation and energy metabolism.

Pathways

AKAP14 functions in PKA-dependent phosphorylation pathways that influence intracellular signaling. In coordination with other proteins it plays roles in the cAMP signaling pathway which regulates cell growth differentiation and apoptosis. AKAP14's interaction with proteins like RII the regulatory subunit of PKA enables the tight modulation of signaling pathways essential for these cellular processes. Another significant pathway involving AKAP14 is the mitotic spindle formation pathway where it ensures centrosome stability and chromosome segregation.

AKAP14's dysfunction is linked to specific mitochondrial-related disorders and certain types of cancer. Disruption in its function can result in cellular signaling imbalance contributing to mitochondrial myopathy due to impaired energy metabolism. Furthermore AKAP14's involvement in centrosome regulation associates it with cancers such as breast cancer where centrosome abnormalities are common. Mutations that affect its interaction with other centrosomal proteins like ASPM could lead to anomalies in cell division promoting oncogenic processes.
