Immunocytochemical analysis of AKAP9 in HeLa cells. Cells were grown on chamber slides and fixed with formaldehyde prior to staining. Cells were probed without (control) or with a AKAP9 monoclonal antibody (ab32679) at a dilution of 1 : 20 overnight at 4 C and incubated with a DyLight-488 conjugated secondary antibody. AKAP9 staining (green) F-Actin staining with Phalloidin (red) and nuclei with DAPI (blue) is shown. Images were taken at 60X magnification.

Immunohistochemistry was performed on normal biopsies of deparaffinized Human pancreas tissue. To expose target proteins heat induced antigen retrieval was performed using 10mM sodium citrate (pH6.0) buffer and microwaved for 8-15 minutes. Following antigen retrieval tissues were blocked in 3% BSA-PBS for 30 minutes at room temperature and probed with a AKAP9 monoclonal antibody (ab32679) at a dilution of 1 : 20 or without primary antibody (negative control) overnight at 4°C in a humidified chamber. Tissues were washed with PBST and endogenous peroxidase activity was quenched with a peroxidase suppressor. Detection was performed using a biotin-conjugated secondary antibody and SA-HRP followed by colorimetric detection using DAB. Tissues were counterstained with hematoxylin and prepped for mounting.

The protein known as A-kinase anchor protein 9 (AKAP9) also called Yotiao is involved in the regulation of protein kinase A (PKA). AKAP9 with a mass of approximately 454 kDa is expressed in multiple tissues including the brain heart and muscle tissue. This protein serves as a scaffolding protein that binds PKA to specific subcellular locations thereby organizing the action of signaling enzymes to their target sites.
Biological function summary

AKAP9 plays an essential role in coordinating cellular signaling events. It acts as a hub in a protein complex that includes PKA protein phosphatase 1 and other signaling molecules facilitating targeted signal transduction. It helps regulate ion channel function and the organization of the cell's cytoskeleton. In neurons AKAP9 modulates synaptic signaling and plasticity which influences neurotransmitter release.

Pathways

AKAP9 interacts with signaling pathways such as the cAMP-dependent pathway and calcium signaling. Through these pathways AKAP9 modulates the activity of several related proteins including PKA itself and other proteins like RyR2 a calcium channel. Its involvement in these pathways indicates important roles in regulating cardiac rhythm and other neuronal functions by targeting enzymes to their specific substrates.

AKAP9 has links to cardiac arrhythmias and neurodegenerative disorders. Long QT syndrome linked to aberrant calcium handling involves AKAP9 through its relationship with RyR2 and other cardiac proteins. In Alzheimer's disease the protein's role in synaptic signaling might connect it to amyloid beta and Tau proteins suggesting its involvement in neuronal dysfunction and disease progression.
