Immunofluorescent analysis of 100% methanol-fixed, 0.1% TritonX-100 permeabilized HT-29 (human colorectal adenocarcinoma epithelial cell) cells labeling ALDH1A3 with ab308526 at 1/100 (5.42 ug/ml) dilution, followed by ab150081 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed antibody at 1/1000 2ug/ml dilution (Green). Confocal image showing cytoplasmic staining in HT-29 cell line, no staining was observed in MCF-7 cell line. Negative control : MCF-7 (PMID : 31197235). The image was taken with a confocal microscope(Leica-Microsystems, TCS SP8). ab195889 Anti-alpha Tubulin mouse monoclonal antibody - Microtubule Marker (Alexa Fluor® 594) was used to counterstain tubulin at 1/200 2.5ug/ml dilution (Red). The Nuclear counterstain was DAPI (Blue). Secondary antibody only control : Secondary antibody is ab150081 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed at 1/1000 2ug/ml dilution.

ALDH1A3 also known as Aldehyde Dehydrogenase 1 Family Member A3 is an enzyme with a molecular mass of approximately 55 kDa. It belongs to the aldehyde dehydrogenase family which is responsible for oxidizing aldehydes into carboxylic acids. ALDH1A3 plays an important role in the metabolism of retinaldehyde into retinoic acid an important biochemical reaction in the body. This enzyme shows high expression in tissues such as the liver kidney and neural tissue.
Biological function summary

The role of ALDH1A3 extends beyond simple metabolic processes. It is involved in the regulation of retinoid signaling influencing various cellular processes such as differentiation and proliferation. ALDH1A3 does not operate in isolation; it is often part of a complex interplay of enzymes that regulate local concentrations of retinoic acid which serves as a signaling molecule influencing gene expression. This regulatory capability underlines its importance in developmental and physiological patterns.

Pathways

ALDH1A3 is essential in the retinoic acid signaling pathway and the metabolic pathway of vitamin A derivatives. It interacts with other ALDH family members such as ALDH1A1 and ALDH1A2 in the synthesis and degradation of retinoic acid. Through these pathways ALDH1A3 also impacts the Wnt and Hedgehog pathways which are important for cellular growth and differentiation.

ALDH1A3 connects with several neurodegenerative and cancer conditions. Studies have indicated a correlation between ALDH1A3 expression and certain types of cancer including breast cancer and glioblastoma. In cancer aberrant ALDH1A3 levels can affect cancer stem cell properties and tumor progression. Additionally the protein interacts with other proteins like ALDH1L1 and ALDH3A1 which might also play roles in tumor metabolism and resistance to chemotherapy.

Catalyzes the NAD-dependent oxidation of aldehyde substrates, such as all-trans-retinal and all-trans-13,14-dihydroretinal, to their corresponding carboxylic acids, all-trans-retinoate and all-trans-13,14-dihydroretinoate, respectively (By similarity) (PubMed : 27759097). High specificity for all-trans-retinal as substrate, can also accept acetaldehyde as substrate in vitro but with lower affinity (PubMed : 27759097). Required for the biosynthesis of normal levels of retinoate in the embryonic ocular and nasal regions; a critical lipid in the embryonic development of the eye and the nasal region (By similarity).
