Flow Cytometry (Intracellular) - Anti-Aldolase+Aldolase C antibody [EPR19355] - BSA and Azide free (AB251296)

This data was developed using ab200049, the same antibody clone in a different buffer formulation.

Intracellular flow cytometric analysis of 4% paraformaldehyde-fixed U-87 MG (Human glioblastoma-astrocytoma epithelial cell line) cells labeling Aldolase + Aldolase C with ab200049 at 1/600 dilution (red) compared with a rabbit monoclonal IgG isotype control (ab172730; black) and an unlabelled control (cells without incubation with primary antibody and secondary antibody; blue). Goat anti rabbit IgG (Alexa Fluor® 488) at 1/500 dilution was used as the secondary antibody.

Product details

ab251296 is the carrier-free version of ab200049.

Patented technology
Our RabMAb® technology is a patented hybridoma-based technology for making rabbit monoclonal antibodies. For details on our patents, please refer to RabMAb® patents.

What are the advantages of a recombinant monoclonal antibody?
This product is a recombinant monoclonal antibody, which offers several advantages including:

  • - High batch-to-batch consistency and reproducibility
  • - Improved sensitivity and specificity
  • - Long-term security of supply
  • - Animal-free batch production

For more information, read more on recombinant antibodies.

Conjugation ready
Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.

Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.

Compatibility
This product is compatible with the Maxpar® Antibody Labeling Kit from Fluidigm, without the need for antibody preparation. Maxpar® is a trademark of Fluidigm Canada Inc.

Supplementary information

Aldolase including Aldolase C is an important enzyme known also as fructose-bisphosphate aldolase. It catalyzes a reversible reaction in the glycolytic pathway where it cleaves fructose 16-bisphosphate into glyceraldehyde 3-phosphate and dihydroxyacetone phosphate. The enzyme's molecular mass is approximately 39 kDa. Aldolase C expresses mainly in the brain and is one of three aldolase isozymes along with Aldolase A and B each showing tissue-specific expression.
Biological function summary

In the glycolysis and gluconeogenesis processes Aldolase C contributes to energy metabolism and the regulation of glucose homeostasis. It can function as a part of a multienzyme complex where glycolytic enzymes associate along with proteins like phosphofructokinase and pyruvate kinase. Aldolase C shares a structural similarity with other aldolase isozymes but its expression pattern reflects a more CNS-oriented role.

Pathways

Quite significant roles for Aldolase C appear in the glycolysis pathway and the gluconeogenesis pathway. The enzyme facilitates the conversion of sugars specifically glucose into energy tying it closely with energy production cycles within the cell. Aldolase C interacts with enzymes like phosphoglycerate mutase and enolase in these pathways which together work to ensure smooth energy transitions and cellular metabolic functions.

Alterations in Aldolase C levels link to certain neurological diseases such as Alzheimer's disease and glioblastoma. In Alzheimer's changes in glucose metabolism may connect to Aldolase C's altered expressional patterns. In glioblastoma high expression suggests a potential role in tumor metabolism. The target's relation to neurodegenerative and cancerous conditions often associates with proteins like tau in Alzheimer's and with signaling proteins in cancer pathways indicating its importance in disease diagnosis and therapy exploration.

Target data

Catalyzes the reversible conversion of beta-D-fructose 1,6-bisphosphate (FBP) into two triose phosphate and plays a key role in glycolysis and gluconeogenesis (PubMed : 14766013). In addition, may also function as scaffolding protein (By similarity).
