Negative immunofluorescence staining of Aggrecan in a section of formalin-fixed paraffin-embedded human liver*. Performed on a Leica BOND. The section was pre-treated using heat mediated antigen retrieval with EDTA (pH9.0) using retrieval settings of 110°C for 40 minutes. The section was then incubated at room temperature for 1 hour with ab318312 at 1/100 dilution. Nuclear DNA was labelled with DAPI (shown in blue). The section was then mounted using Dako Fluorescence Mounting Medium ®. Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8). For other IHC staining systems (automated and non-automated), customers should optimize variable parameters such as antigen retrieval conditions, antibody concentrations and incubation times. *Tissue obtained from the Human Research Tissue Bank, supported by the NIHR Cambridge Biomedical Research Centre.

Specificity

This antibody detects recombinant human Aggrecan by Western Blotting. We have been unable to detect endogenous protein in WB with this antibody. However, there are many citations indicating this antibody is able to detect Aggrecan in endogenous sample lysates.

Aggrecan also known as aggrecan core protein or chondroitin sulfate proteoglycan 1 is a large proteoglycan vital for cartilage structure and function. It has a mass of approximately 250 kDa. Aggrecan is expressed mostly in cartilage regions such as articular cartilage but it might also be found in other connective tissues. The structure of aggrecan consists of a core protein that attaches to glycosaminoglycan chains allowing it to create hydrated gel structures essential for the resilience and load-bearing properties of cartilage.
Biological function summary

Aggrecan plays a significant role in maintaining the structural integrity of cartilage by providing resistance to compression. This proteoglycan is a central component of the extracellular matrix forming large aggregates with hyaluronic acid and link proteins. These aggregates contribute to the cartilage's osmotic properties helping resist compression during joint movement. Aggrecan's structural properties make it necessary for normal joint function and health.

Pathways

Aggrecan interacts within the cartilage development and maintenance frameworks. Specifically it participates in the regulation of the ECM-receptor interaction pathway and the proteoglycans in cancer pathway. Aggrecan functions alongside proteins such as type II collagen to ensure biomechanical properties of the cartilage. Additionally its enzymatic breakdown by aggrecanases like ADAMTS can impact these pathways leading to various physiological effects.

Aggrecan is closely associated with osteoarthritis and intervertebral disc degeneration. In osteoarthritis the loss of aggrecan through enzymatic degradation leads to reduced joint function and increased pain. The breakdown involves aggrecanases and other matrix metalloproteinases resulting in cartilage degradation. Diseases such as intervertebral disc degeneration also link to the reduced presence or altered structure of aggrecan impacting spine health and mobility.
