Immunocytochemistry/ Immunofluorescence - Alexa Fluor® 488 Anti-BrdU antibody [BU1/75 (ICR1)] (AB220074)

ab220074 staining BrdU in HeLa cells. Untreated and BrdU treated (10uM for 24 hours) cells were fixed with 100% methanol (5 min) and then subjected to acid hydrolysis using 2M HCL in 0.1% PBS-Tween for 30 minutes at room temperature to denature the DNA. They were then permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab220074 at 1/100 dilution (shown in green) and ab195889, Mouse monoclonal to alpha Tubulin (Alexa Fluor® 594), at 1/250 dilution (shown in red). Nuclear DNA was labelled with DAPI (shown in blue).

Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

This product also gave a positive signal under the same testing conditions in HeLa cells fixed with 4% formaldehyde (10 min).

Specificity

This antibody reacts with BrdU in single stranded DNA, BrdU attached to a protein carrier or free BrdU. It detects nucleated cells in S-Phase which have had BrdU incorporated into their DNA. Also reacts with chlorodeoxyuridine but with reduced staining. The antibody does not react with thymidine. It has been reported in the literature that this antibody clone cross-reacts with Edu (PMID: 23272138) and some customers reported that it cross reacts with IdU.

Supplementary information

BrdU short for Bromodeoxyuridine is a synthetic analog of the nucleoside thymidine. It substitutes thymidine in newly synthesized DNA allowing for the tracking of cellular proliferation. Bromodeoxyuridine integrates into the DNA of proliferating cells which can then be detected using specific anti-BrdU antibodies. These antibodies are valuable tools in various assays such as BrdU staining and BrdU proliferation assay. BrdU has a molecular weight of approximately 315.13 g/mol. It is typically utilized in tissues or cell lines where active DNA synthesis and cellular proliferation are occurring.
Biological function summary

Bromodeoxyuridine acts as a marker for identifying cells in the S-phase of the cell cycle. During this phase DNA synthesis occurs and BrdU incorporates into the replicating DNA strand. When used in combination with anti-BrdU antibodies it enables researchers to identify and quantify dividing cells. This capability makes BrdU incorporation a widely accepted method to study cell proliferation in various biological samples. It functions independently without forming larger complexes.

Pathways

Bromodeoxyuridine integration highlights its role in DNA replication and repair pathways. It helps in the study of cell growth regulation pathways where proteins like PCNA (Proliferating Cell Nuclear Antigen) are involved. BrdU pairing with anti-BrdU antibodies allows assessment of DNA synthesis therefore aiding in the investigation of critical pathways like the cell cycle regulation pathway. In this context BrdU kits provide a consistent method for assessing dynamic changes in proliferative states.

Bromodeoxyuridine is useful in cancer research and neurological disorders like Alzheimer's disease. Through its ability to track cell proliferation researchers can explore cancerous tissue's growth patterns elucidating pathways and factors driving tumor expansion. BrdU assays contribute to understanding the aberrant cellular proliferation seen in tumors. For Alzheimer's disease BrdU aid in investigating neurogenesis rates particularly in regions affected by the disorder. Researchers often connect BrdU results with proteins like tau and amyloid-beta which play roles in these conditions.
