Immunohistochemistry (Frozen sections) - Alexa Fluor® 488 Anti-CD11b + CD11c antibody [OX42] (AB216355)
IHC image of CD11b + CD11c staining in a section of frozen normal rat brain.
The section was fixed using 10% formaldehyde in 1XPBS for 10 minutes. No antigen retrieval step was performed prior to staining. Non-specific protein-protein interactions were then blocked in TBS containing 0.025% (v/v) Triton X-100, 0.3M (w/v) glycine and 1% (w/v) BSA for 1h at room temperature. The section was then incubated overnight at +4°C in TBS containing 0.025% (v/v) Triton X-100 and 1% (w/v) BSA with ab216355 at 1/100 dilution (shown in green) and counterstained using ab195884, Rat monoclonal to Tubulin (Alexa Fluor® 647), at 1/250 dilution (shown in red). Nuclear DNA was labelled with DAPI (shown in blue). The section was then mounted using Fluoromount®.
Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).
For other IHC staining systems (automated and non-automated), customers should optimize variable parameters such as antigen retrieval conditions, antibody concentrations and incubation times.

Product details
PMID: 7868861. The antigen is temperature sensitive; we may recommend following the conditions as recommended in this publication. As per publication low temperature paraffin, using 0.5 to 2% paraformaldehyde fixation instead of 4% or using periodate-lysine-paraformaldehyde (PLP) + 0.25% PFA and 0.1% glutaraldehyde might be helpful in detection. In case of any question please do not hesitate to contact technical team at technical@abcam.com.
The principal use of this antibody is in the study of functional heterogeneity of macrophages; it may be used to follow macrophage differentiation and to investigate the function of the CD11b + CD11c equivalent antigen.

Biological function summary
Integrins αM (CD11b) and αX (CD11c) facilitate key processes such as adhesion and migration of leukocytes. Both CD11b and CD11c pair with CD18 to form the Mac-1 (Macrophage-1 antigen also known as CR3) and p15095 complexes respectively. These complexes participate in both innate and adaptive immune responses by binding to various ligands like ICAM-1 and fibrinogen. CD11b/CD18 (Mac-1) additionally plays an essential role in phagocytosis degranulation and respiratory burst important activities for pathogen clearance.
Pathways
These integrins engage in critical signaling pathways including the NF-kappaB and MAPK pathways. They regulate the inflammatory response and cell survival. CD11b interacts closely with other proteins like ICAM-1 and C3b which are involved in these pathways. CD11c also contributes to similar pathways although it plays a more specialized role in dendritic cells impacting antigen presentation and immune regulation.
