Flow Cytometry - Alexa Fluor® 488 Anti-CD41 antibody [M148] (AB233053)

Overlay histogram showing left HEL92.1.7 positive cells and right negative Jurkat cells stained with ab233053 (red line). The cells were incubated in 1x PBS containing 10 % normal goat serum to block non-specific protein-protein interactions followed by the antibody (ab233053) (1x106 in 100 μl at 1 μg/ml (1/500)) for 30 min on ice. Isotype control antibody (black line) was mouse IgG2a kappa; Alexa Fluor® 488 used at the same concentration and conditions as the primary antibody. Unlabelled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 50 mW Blue laser (488nm) and 530/30 bandpass filter.

This antibody binds to some medulloblastoma and neuroblastoma cells and rhabdomyosarcoma and certain other solid tumours. No binding observed to marrow constituents other than platelets and megakaryocytes.

Target data

Integrin alpha-IIb/beta-3 is a receptor for fibronectin, fibrinogen, plasminogen, prothrombin, thrombospondin and vitronectin. It recognizes the sequence R-G-D in a wide array of ligands. It recognizes the sequence H-H-L-G-G-G-A-K-Q-A-G-D-V in fibrinogen gamma chain (By similarity). Following activation integrin alpha-IIb/beta-3 brings about platelet/platelet interaction through binding of soluble fibrinogen (PubMed : 9111081). This step leads to rapid platelet aggregation which physically plugs ruptured endothelial cell surface (By similarity).
