Flow Cytometry - Alexa Fluor® 488 Anti-CD45RA antibody [MRC OX-33] (AB256263)
Lewis rat splenocytes stained with ab256263 (right) or mouse IgG1κ Alexa Fluor® 488 isotype (left). Lewis rat splenocytes were incubated for 30 min on ice in 1x PBS / 10 % rat serum to block FC receptors and non-specific protein-protein interaction followed by the antibody (ab256263) or mouse IgG1κ Alexa Fluor® 488 isotype (1x 106 in 100 μl at 0.2 μg/ml (1/2500)) for 30 min on ice. The cells were simultaneously stained with CD3.
Acquisition of >30,000 events were collected using a 50 mW Blue laser (488nm) and 530/30 bandpass filter. Events were gated on viable lymphocytes.

Product details
Spleen cells from immunised BALB/c mice were fused with cells of the NSO/U myeloma cell line. This clone has been described reacting with paraffin embedded material following PLP fixation (see Whiteland et al., 1995). It only labels B cells among thoracic duct lymphocytes, with little labelling in bone marrow and none on thymocytes (Barclay et al., 1987).

Biological function summary
CD45RA plays a significant role in regulating T-cell receptor (TCR) signaling. By dephosphorylating specific phosphotyrosine residues on substrates CD45RA modulates signal transduction pathways important for T-cell activation and differentiation. This protein does not act alone but is part of a regulatory network that influences immune responses and cellular development through its interaction with other cell surface receptors and intracellular kinases.
Pathways
CD45RA modulation is integral to T-cell development and activation processes. It is an important participant in the T-cell receptor signaling pathway and the Jak-STAT signaling pathway. In these pathways CD45 interacts with proteins such as Lck and Fyn which are essential for propagating receptor-mediated signaling events in T-cells. Effective signal regulation through CD45RA is necessary to ensure appropriate immune responses.
