Immunohistochemical analysis of paraffin-embedded human colon tissue labeling Cytokeratin 20 with ab275988 at 1/200 (5.0 μg/ml) dilution (Green).

Positive staining on human colon.

The section was pre-treated using heat mediated antigen retrieval with Tris/EDTA buffer (pH 9.0) 110℃ for 20 mins. Non-specific protein-protein interactions were then blocked in TBS containing 0.025% (v/v) Triton X-100, 0.3M (w/v) glycine and 1% (w/v) BSA for 1h at room temperature. The section was then incubated overnight at +4°C in TBS containing 0.025% (v/v) Triton X-100 and 1% (w/v) BSA with ab275988 at 1/200 dilution (shown in green). Nuclear DNA was labeled with DAPI (shown in blue). The section was then mounted using Fluoromount ®.

Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

Cytokeratin 20 often known as CK20 KRT20 or keratin 20 is a type of intermediate filament protein that is highly specific to epithelial cells. This cytoskeletal protein has a molecular weight of approximately 46.5 kDa. Cytokeratin 20 is primarily found in the gastric and intestinal epithelium urothelium and Merkel cells. Its function as part of the cytoskeleton provides structural integrity and strength to cells maintaining their shape and resilience against mechanical stress.
Biological function summary

Cytokeratin 20 is essential in cellular differentiation and maintenance within epithelial tissues. It plays an important role in the structural framework of epithelial cells by integrating into the cytoskeletal network which includes other keratins.
