Activity summary

The Estrogen Inducible Protein pS2 also known as Trefoil Factor 1 (TFF1) is a small protein with a mass of about 6.6 kDa. It is mainly expressed in the mucosa of the stomach and other tissues responsive to estrogen. pS2 functions mechanically as part of the trefoil factor family known for its role in mucosal protection and repair. pS2 has a distinctive three-loop structure which is key to its interaction with other molecules and substrates in the cellular environment.

Biological function summary

This trefoil factor plays a significant role in the process of epithelial regeneration and maintaining gastrointestinal tract integrity. It is not known to be part of a complex but works through interaction with various growth factors and cytokines. Its expression is regulated by estrogen making pS2 significant in estrogen-responsive tissues. These interactions contribute to maintaining the stability of epithelial lining aiding in cellular migration and wound healing.

Pathways

PS2 influences cellular signaling involved in the gastric mucosal defense mainly interacting with the Epidermal Growth Factor (EGF) pathway. It modulates the pathway's effects on cell proliferation and migration. Its role is linked to the mitigation of tissue damage indicating a connection with other proteins like the EGF receptor contributing to the repair and defense mechanisms of epithelial tissues.

Associated diseases and disorders

PS2 has gained attention in breast cancer research. Its expression correlates with hormone receptor-positive breast cancer making it a potential biomarker for the disease. Additionally aberrant expression levels of pS2 might relate to gastric cancer where it plays a role in tumor progression and response to therapy. pS2 is closely connected to estrogen receptor proteins which guide its involvement in these cancers influencing prognosis and treatment strategies.

    Overlay histogram showing MCF7 cells stained with ab200799 (red line). The cells were fixed with 4% formaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab200799, 1/50 dilution) for 30 min at 22°C. Isotype control antibody (black line) was rabbit monoclonal IgG [EPR25A] Alexa Fluor® 488 (Alexa Fluor® 488 Rabbit IgG, monoclonal [EPR25A] - Isotype Control ab199091) used at the same concentration and conditions as the primary antibody. Unlabelled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter. This antibody gave a positive signal in MCF7 cells fixed with 80% methanol (5 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions.

    ab200799 staining Estrogen Inducible Protein pS2 in MCF7 cells. The cells were fixed with 4% formaldehyde (10min), permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab200799 at 1/200 dilution (shown in green) and Alexa Fluor® 594 Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker ab195889, Mouse monoclonal to alpha Tubulin (Alexa Fluor® 594), at 2μg/ml (shown in red). Nuclear DNA was labelled with DAPI (shown in blue).
