ICAM-3 also known as CD50 or Intercellular Adhesion Molecule 3 is a cell surface glycoprotein. It weighs approximately 120 kilodaltons. ICAM-3 is expressed majorly on leukocytes including lymphocytes monocytes and granulocytes. It is a member of the immunoglobulin superfamily that participates in cell-cell interaction particularly in immune response. By engaging with integrin receptors mainly LFA-1 ICAM-3 plays a role in leukocyte adhesion and signaling.
Biological function summary

ICAM-3 contributes to the immune system’s functionality by mediating intercellular adhesion and cellular signaling. It acts as a ligand for the integrin LFA-1 found on leukocytes which facilitates the initial immune response. ICAM-3 is not a part of a larger complex but works closely with LFA-1 in homing and recruitment of immune cells to inflammation sites. Its interaction with LFA-1 also supports the formation of the immunological synapse an important step in T-cell activation and immune signaling.

Pathways

ICAM-3 contributes to both the immune response and leukocyte extravasation pathways. The interaction with LFA-1 places it within the context of immune cell signaling pathways impacting integrin-mediated signaling. Another pathway involving ICAM-3 is the cell adhesion pathway which is essential for maintaining proper immune surveillance and response. It collaborates with proteins like ICAM-1 in these pathways to ensure effective immune activity.

ICAM-3 is associated with conditions such as inflammatory diseases and certain autoimmune disorders. Its expression levels and binding interactions can affect the severity and progression of such diseases. In autoimmune diseases ICAM-3 interaction with LFA-1 might contribute to the aberrant immune activation observed. Additionally studies have highlighted its role in infections where viruses exploit ICAM proteins to enter or spread showing its importance in pathogenesis-related conditions.

ICAM proteins are ligands for the leukocyte adhesion protein LFA-1 (integrin alpha-L/beta-2) (PubMed : 1448173). ICAM3 is also a ligand for integrin alpha-D/beta-2. In association with integrin alpha-L/beta-2, contributes to apoptotic neutrophil phagocytosis by macrophages (PubMed : 23775590).
