ab202944 staining LYRIC/AEG1 in A673 cells. The cells were fixed with 4% formaldehyde (10 min), permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab202944 at a 1/100 dilution (shown in green) and ab202272, Rabbit monoclonal to alpha Tubulin (Alexa Fluor® 594), at a 1/250 dilution (shown in red). Nuclear DNA was labelled with DAPI (shown in blue).

Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

This product also gave a positive signal under the same testing conditions in A673 cells fixed with 100% methanol (5 min).

LYRIC/AEG1 also known as Astrocyte Elevated Gene-1 (AEG-1) is a protein with a molecular weight of approximately 60 kDa. This protein is expressed in several tissues with notable expression in the brain liver and other organs. LYRIC/AEG1 is encoded by the MTDH gene and plays a mechanical role in regulating processes such as gene expression and cell survival. It localizes to the endoplasmic reticulum the perinuclear region and also the cell nucleus indicating a multifaceted role in cellular dynamics.

Down-regulates SLC1A2/EAAT2 promoter activity when expressed ectopically. Activates the nuclear factor kappa-B (NF-kappa-B) transcription factor. Promotes anchorage-independent growth of immortalized melanocytes and astrocytes which is a key component in tumor cell expansion.
