Flow cytometric analysis of 4% paraformaldehyde fixed 90% methanol permeabilized THP-1 (human monocytic leukemia monocyte) treated with 100ng/ml PMA for 56 hours, add 1 μg/ml LPS for another 16 hours (Red) / Untreated control (Green) cells labelling MIP-1 alpha/CCL3 + CCL3L1 with ab307620 at 1/50 dilution (1μg) (Red) and Green (Red) compared with a Rabbit IgG monoclonal [EPR25A] - Isotype Control (Alexa Fluor® 488) (ab199091) (Black) isotype control and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (Blue).

Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% TritonX-100 permeabilized THP-1 (human monocytic leukemia monocyte) cells labelling MIP-1 alpha/CCL3 + CCL3L1 with ab307620 at 1/250 (2.0 μg/ml) dilution. Confocal image showing cytoplasmic staining in THP-1 treated with Phorbol-12-myristate-13-acetate (100ng/ml) for 56 h, followed by adding Lipopolysaccharide (1μg/ml) for a further 16 h. Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8). ab195889 Anti-alpha Tubulin mouse monoclonal antibody - Microtubule Marker (Alexa Fluor® 594) was used to counterstain tubulin at 1/200 2.5ug/ml dilution (Red). The Nuclear counterstain was DAPI (Blue).

MIP-1 alpha also known as CCL3 and CCL3L1 is a chemokine with a core protein mass around 8 kDa. It is primarily expressed in activated T cells monocytes and lymphocytes. CCL3 plays a role in chemotaxis by binding to receptors CCR1 and CCR5 on the surface of target cells. This interaction leads to cell signaling events that control the migration and activation of specific immune cells to sites of inflammation or infection.
Biological function summary

The molecule acts as an important mediator in the immune response by recruiting and activating neutrophils eosinophils and other leukocytes. It supports inflammatory responses particularly in allergic reactions and autoimmunity. While not a part of a traditional complex MIP-1 alpha directly interacts with its receptors to initiate immune signaling pathways. Its expression can be upregulated by pro-inflammatory cytokines thereby enhancing its biological effects during immune system activation.

Pathways

MIP-1 alpha is closely intertwined in the chemokine signaling pathway and plays an important role in the inflammatory response pathway. Within these pathways MIP-1 alpha coordinates with proteins like CCL5 (RANTES) and interleukin-8 facilitating the recruitment of immune cells. Its function provides a pivotal control point in the migration and activation of these cells influencing the overall immune response.

MIP-1 alpha is strongly associated with rheumatoid arthritis and human immunodeficiency virus (HIV) infection. The chemokine contributes to the inflammatory milieu present in rheumatoid arthritis by promoting the recruitment of inflammatory cells to the synovial membrane. In HIV MIP-1 alpha competes with the virus for binding to CCR5 receptors influencing disease progression. It shares its target receptor CCR5 with proteins such as CCL4 (MIP-1 beta) highlighting its importance in the context of viral diseases and autoimmune conditions.
