Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Alexa Fluor® 488 Anti-Serpin A5 antibody [EPR23337-112] (AB275185)
Immunofluorescence staining of Serpin A5 staining in a section of formalin-fixed paraffin-embedded human normal kidney*.
Performed on a Leica BOND. The section was pre-treated using heat mediated antigen retrieval with EDTA (pH9.0) using retrieval settings of 110°C for 20 minutes. The section was then incubated at room temperature for 1 hour with ab275185 at 1/100 dilution (shown in green) and counterstained using ab190573, Rabbit anti-Tubulin (A647)(1/250) at 1/250 dilution (shown in red). Nuclear DNA was labelled with DAPI (shown in blue). The section was then mounted using Dako Fluorescence Mounting Medium ®.
Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).
For other IHC staining systems (automated and non-automated), customers should optimize variable parameters such as antigen retrieval conditions, antibody concentrations and incubation times.

Serpin A5 does not form part of larger protein complexes but exerts its function through direct interactions. Its modulatory action impacts processes such as cell migration and tumor invasion highlighting its role in broader cellular environments.
Pathways
Serpin A5 is an important regulator in the coagulation cascade and fibrinolytic pathways. It acts to modulate the balance between coagulation and fibrinolysis critical for maintaining hemostasis. This protein is related to other serpins such as Antithrombin III and Plasminogen activator inhibitor-1 through its regulatory actions in these pathways. The inhibition of proteases by Serpin A5 ensures proper activation and termination of these biological processes which are essential for vascular integrity.
