Flow Cytometry (Intracellular) - Alexa Fluor® 488 Anti-TCF-4 antibody [NCI-R159-6] (AB246762)

Overlay histogram showing SH-SY5Y cells stained with ab246762 (red line). The cells were fixed with 4 % formaldehyde (10 min) and then permeabilized with 0.1 % PBS-Triton X-100 for 15 min. The cells were then incubated in 1x PBS containing 10 % normal goat serum to block non-specific protein-protein interaction followed by the antibody (ab246762) (1x 106 in 100μl at 1 μg/ml (1/500)) for 30 min at 22°C.

Isotype control antibody (black line) was Rabbit IgG monoclonal A488 used at the same concentration and conditions as the primary antibody. Unlabelld sample (blue line) was also used as a control.

Acquisition of >5,000 events were collected using a 50 mW Blue laser (488nm) and 530/30 bandpass filter.

Target data

Transcription factor that binds to the immunoglobulin enhancer Mu-E5/KE5-motif. Involved in the initiation of neuronal differentiation. Activates transcription by binding to the E box (5'-CANNTG-3'). Binds to the E-box present in the somatostatin receptor 2 initiator element (SSTR2-INR) to activate transcription (By similarity). Preferentially binds to either 5'-ACANNTGT-3' or 5'-CCANNTGG-3'.
