Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Alexa Fluor® 555 Anti-CNPase antibody [11-5B] (AB317592)

Negative immunofluorescence staining of CNPase in a section of formalin-fixed paraffin-embedded mouse skeletal muscle*.

Performed on a Leica BOND™. The section was pre-treated using heat mediated antigen retrieval with EDTA (pH9.0) using retrieval settings of 110°C for 40 minutes. The section was then incubated at room temperature for 1 hour with ab317592 at 1/100 dilution. Nuclear DNA was labelled with DAPI (shown in blue). The section was then mounted using Dako Fluorescence Mounting Medium®.

Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

For other IHC staining systems (automated and non-automated), customers should optimize variable parameters such as antigen retrieval conditions, antibody concentrations and incubation times.

Immunocytochemistry/ Immunofluorescence - Alexa Fluor® 555 Anti-CNPase antibody [11-5B] (AB317592)

ab317592 staining CNPase in primary hippocampal rat neurons/glia, (obtained from Neuromics, cat. no. PC35101), DIV14. The cells were fixed with 4% formaldehyde (10 min), permeabilised with 0.1% Tween for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab317592 at 5 μg/ml (shown in green). ab195884, Rat monoclonal to Tubulin (Alexa Fluor® 647) (shown in magenta) was used at 2 μg/ml for structural counterstaining.

Image was acquired with a confocal microscope (Leica-Microsystems TCS SP8) and a single confocal section is shown.

This product also work with 100% methanol (5 min) fixation under the same testing conditions.

Target data

Myelin-associated enzyme that catalyzes the phosphodiester hydrolysis of 2',3'-cyclic nucleotides to 2'-nucleotides. In the mitochondria, regulates the functioning of the mitochondrial permeability transition pore (mPTP), and thus is involved in the mechanisms of cell death, both apoptosis and necrosis. Acts as an antiviral factor by suppressing the assembly of SARS-CoV-2 virions (PubMed : 38117830).. Acts as an antiviral factor by suppressing the assembly of HIV-1 virions through direct interaction with GAG.
