Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Alexa Fluor® 594 Anti-Estrogen Receptor alpha antibody [EPR4097] (AB207046)
Immunofluorescence staining of Estrogen Receptor alpha staining in a section of formalin-fixed paraffin-embedded human breast ductal carcinoma*.
Performed on a Leica BOND. The section was pre-treated using heat mediated antigen retrieval with EDTA (pH9.0) using retrieval settings of 110°C for 20 minutes. The section was then incubated at room temperature for 1 hour with ab207046 at 1/100 dilution (shown in red) and counterstained using ab195887, Mouse monoclonal to alpha Tubulin (Alexa Fluor® 488), at 1/250 dilution (shown in green). Nuclear DNA was labelled with DAPI (shown in blue). The section was then mounted using Dako Fluorescence Mounting Medium®.
Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).
For other IHC staining systems (automated and non-automated), customers should optimize variable parameters such as antigen retrieval conditions, antibody concentrations and incubation times.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Alexa Fluor® 594 Anti-Estrogen Receptor alpha antibody [EPR4097] (AB207046)
Immunohistochemistry analysis (Formalin/PFA-fixed paraffin-embedded sections) of formaldehyde-fixed human breast tissue permeabilized with 0.05% Tween 20. Stained with ab207046 at 1/100 dilution. Blocking was done with Sea Block for 30 minutes at 22°C. The sample was incubated with the primary antibody and Sea Block for 16 hours at 4°C. Antigen retrieval method was heat mediated, ab64236 100x Citrate Buffer pH 6.0.

Biological function summary
ERα plays a significant role in the regulation of estrogen signaling. Estrogens binding to ERα activate the receptor which can form a homodimer or heterodimer complex with other proteins like coactivators or corepressors. This complex then modulates the transcription of genes involved in cell growth proliferation and differentiation. ERα is closely linked with processes like reproductive tissue development and maintenance.
Pathways
ERα is involved in the estrogen signaling pathway and the cell cycle regulation pathway. In the estrogen signaling pathway ERα works together with proteins such as coactivators which enhance gene transcription and corepressors which can inhibit transcription. In the context of cell cycle regulation ERα's interactions with other cell cycle proteins help control cell division and proliferation linking ERα activity to the progression through different stages of the cell cycle.
