ETV2 also known as ETS variant transcription factor 2 functions as a transcription factor important in the regulation of gene expression involved with endothelial lineage differentiation. This protein has a mass of approximately 38 kDa. ETV2 is mainly expressed in embryonic vascular endothelial cells as well as in adult tissues like the heart and lungs. Some studies also refer to it by the alternate name ER71. High expression levels in these key areas support its critical roles in vasculature development and function.
Biological function summary

ETV2 regulates endothelial cell development and vasculogenesis. It acts as an important switch for endothelial cell fate essentially directing stem cells towards an endothelial lineage. ETV2 often operates within a larger protein complex interacting with other transcription factors necessary for vascular differentiation. Its role indicates a pivotal function in blood vessel formation and remodeling greatly influencing vascular health.

Pathways

ETV2 is a part of the VEGF (vascular endothelial growth factor) signaling pathway and the NOTCH signaling pathway. In the VEGF pathway ETV2 directly influences genes responsible for angiogenesis the process of forming new blood vessels. The NOTCH pathway involves ETV2 in the differentiation and proliferation of endothelial cells. ETV2 interacts with proteins such as VEGFA and NOTCH1 both significant contributors to vascular biology and embryonic development.

ETV2 relates to cardiovascular diseases and certain cancers. Dysregulation of ETV2 expression can contribute to impaired blood vessel formation playing a part in conditions like atherosclerosis and peripheral artery disease. The link between ETV2 and cancers such as breast cancer reveals its involvement in tumor angiogenesis as tumors harness ETV2 to recruit blood vessels promoting their growth. ETV2 also connects to the protein HIF1A which plays a critical role in cancer angiogenesis under low oxygen conditions.
