Immunocytochemistry/ Immunofluorescence - Alexa Fluor® 647 Anti-CD3 zeta (phospho Y142) antibody [EP265(2)Y] (AB310016)
Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% TritonX-100 permeabilized Jurkat (human T cell leukemia T lymphocyte from peripheral blood) cells labelling CD3 zeta (phospho Y142) with ab310016 at 1/50 dilution (10.0 µg/ml). ab195887, Anti-alpha Tubulin mouse monoclonal antibody - Microtubule Marker (Alexa Fluor® 488) was used at 1/200 dilution (2.5 µg/ml) for Tubulin counter stanining (Green). DAPI was used as nuclear counter stain. Confocal image showing increased cytoplasmic staining in Jurkat cells treated with pervanadate (1 mM) for 30 min. Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

Flow cytometric analysis of 4% paraformaldehyde fixed, 90% methanol permeabilized Jurkat (human T cell leukemia T lymphocyte from peripheral blood) treated with 1mM Pervanadate for 30min (Red) / Untreated Jurkat (Green) cells labelling CD3 zeta (phospho Y142) with ab310016 at 1/500 dilution (0.1 ug) /(Red and Green) compared with a Rabbit IgG monoclonal [EPR25A] - Isotype Control (Alexa Fluor® 647) (ab199093) / (Black) isotype control and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (Blue).
